Abstract
This study aimed to optimize the regeneration and transformation systems for eight durum wheat genotypes obtained from ICARDA through a twinning agreement with AGERI as compared to the Egyptian durum wheat cultivars Sohag3 and BeniSweif3, respectively. To achieve this goal, three different experiments were conducted. In first experiment two different growth regulators; TDZ with concentration (0.2 mg/l) and KN+NAA with concentration (1 mg/l each), were compared as different growth regulators. In the second experiment, regeneration efficiencies of calli induced from different genotypes after biolistic bombardment was measured and the amenability of wheat genotypes to transform via the biolistic device was detected, both of callus induction and regeneration media contained either 0 mg/l or 3 mg/l bialaphos. While, in third experiment, the biolistic gun conditions were optimized.
