Abstract

Fusarium oxysporum is a common soil born plant pathogen with Egypt and worldwide distribution .Wilt is considered as one of the major factors for low productivity of cucumber (Cicer arietinum). Due to the importance of this disease , four Fusarium isolates collected from different soil types were identified and screened for patogenicity and genetic variation using RAPD-DNA markers. Disease severity and variations in reaction symptoms were recorded for all isolates. The F. oxysporum isolate RAPD profiles were not typical, all four showing close similarity after the development of a phyllogenetic tree. The RAPD-PCR may be valuable in monitoring populations and characterize different pathotypes of F. oxysporum .Also, clustering analysis based on RAPD fingerprint data revealed two distinct groups within F. oxysporum which often correspond to the origin of isolate. Genetic similarity between each of the isolates was calculated and results indicate that there was little genetic variability among the isolates collected from the different locations
