Abstract

INTRODUCTION:

Acute kidney injury (AKI) is a syndrome of rapidly declining renal function.
Stem cell is the origin of an organism's life Several studies demonstrated that the administration of in vitro expanded mesenchymal stem cells (MSCs) improves acute kidney injury. However, the mechanisms involved in renal regeneration induced by MSCs remain controversial.
The administration of conditioned medium(CM) from MSCs may mimic the beneficial effects of the MSC administration. Besides soluble factors, microvesicles (MVs) derived from MSCs contribute to kidney repair in experimentally induced AKI.
AIM OF THE WORK
To evaluate the possible beneficial effect of mesenchymal stem cells, their conditioned medium and mesenchymal stem cells derived microvesicles in treatment of acute kidney injury.
METHODS:
Animals were divided into 5 groups,(each contained 10 rats) as follows:
Group I(control group) : rats were given saline
Group II (affected group): They were given glycerol (8 ml/kg/50%) intramuscularly distributed equally in both hind limbs as a single dose to induce acute kidney injury.
Group III(stem cell group): They were given mesenchymal stem cells ( MSCs ) 2million cell/kg by intravenous route in tail vein 3 days after being given glycerol (8 ml/kg/50%) intramuscularly distributed equally in both hind limbs as a single dose.
Group IV(Conditioned medium group): They were given the conditioned medium(CM) by intravenous route in tail vein 3days after being given glycerol (8 ml/kg/50%) intramuscularly distributed equally in both hind limbs as a single dose.
Group V( microvesicles group): They were given microvesicles by intravenous route in tail vein 3days after being given glycerol (8 ml/kg/50%) intramuscularly distributed equally in both hind limbs as a single dose.
